Fixation in cytology:-
After collection of specimen make smear in clean slide and fixed by 95% alcohol while the preparation it wet to avoid drying and shrinkageof cells.
The pest using equal part of Ether and Alcohol, but not large amount Because it flammable.
بعد جمع العينه تعمل مسحة علي الشريحة الزجاجية وتثبت باستخدام 95% ايثر لتجنب الجفاف والانكماش للخلايا ونستخدم في التثبيت خليط من الايثر والكحول ولا نستخدم الكحول بكمية كبيرة لانه سريع الاشتعال.
Staining in cytology:-
Using Papa nicolous stain(Pap stain) which contain:-
-1Harris alum haematoxyline
3%glycial actic acid to staining the Nuclous
2-orang G6(OG6) contain:-
Phosphor Tongestic acid it act as mordant
3- Eosin Azuor50 or 60 it contain:-
1-light green
2-Bis merk brown
3-Eosin
4-phospho tongestic Acid
5-lithium carbonate
Result:-
Superficial cell:cytoplasm_ pink in colour
Intermediate cell parabasal cell:blue
Nucloucs:blue

Examenation and Identification by 3 method
1-Maturation Index
Count last 200 squamaous cell the degree of cell dependent to 1-the morphology 2- Staining reaction 3-type of cell (inter mediate-parabasal)
The result is expressed as( presentage %) high or low of Estrogen activity .
2-Eosinophilic Index:-
Count 100 Cell and detect How many cells of 100 are Eosinophilic.
In Causes inflammation the cytoplasm with Eosin.
3-karyo pyknotic Index(KPF):-
Pyknotic means :Nuclous without details .
Count last 200 squamous Cell .
Pyknotic –Hyper chromatic staining reaction not more than 5Mm in diameter.
