why we used spectrophotometer blank???

السلام وعليكم ورحمة الله وبركاته
عندي سؤال وأرجوا الافادة الشافية الكافية لو سمحتم
لماذا يلزم فحص الreagent على جهاز spectrophotometer في كل فحص علما بان ورقة الفحص المكتوبة على ال reagent يكون معروفة قيمته يعني (100 في فحص السكر ...مثلا ، ولماذا اختير وكيف هذا الرقم؟)
2 - كما انه لماذا يلزم بعض الفحوصات ان يتم تصفير الجهاز على هواء أو ماء مقطر ايضا
ولكم جزيل الشكر افيدونا بسرعة اذا ممكن

السبب هو التالي:

Because other compounds in a solution (or the solvent itself) may absorb the same wavelengths as the compound being analyzed, we compare the absorbance of our test solution to a reference blank. Ideally, the reference blank should contain everything found in the sample solution except the substance you are trying to analyze or measure. For instance, in today’s lab exercise you will be measuring the absorbance of a dye, bromphenol blue that was dissolved in water The reference blank in this case would be water alone.

بكل بساطة البلانك دايماً له قراءةً (حتى لو كانت قيمتها صغيرة جدا) في الspectrophotometer تطرح من قيمتها قراءة باقي العينات حتى لاتؤثر على النتائج...
