    السلام عليكم و رحمه الله ........

    هذا الموضوع يتكلم عن تقنية ليست بالحديثة ولا بالقديمه و لكنها المفضله و المطبقة الان في دراسه تكثير و تكبير الدي ان اي ....


    Polymerase Chain Reaction (PCR), invented by Kary B. Mullis, at the Cetus Corporation, who was awarded the 1993 Nobel Prize for chemistry for PCR, is a technique to exponentially amplify in vitro a small quantity of a specific nucleotide sequence in the presence of template sequence, two oligonucleotide primers that hybridize to opposite strands and flank the region of interest in the target DNA, a thermostable (taq) DNA polymerase. The reaction is cycled involving template denaturation, primer annealing, and the extension of the annealed primers by DNA polymerase until enough copies are made for further analysis.

    Polymerase Chain Reaction






    Polymerase Chain Reaction (PCR) is a technique for reproducing specific DNA sequences in vitro.

    The process is carried out within a machine (see photo), and it can produce millions or
    illions of copies of a piece of DNA in a few hours.



    The sequence of PCR involves the following steps:

    A: The DNA to be reproduced is heated to separate the two template strands.
    B: Two primers which are complimentary to the region to be amplified are added.
    A heat-stable DNA polymerase enzyme is also added. The enzyme catalyses the extension of the primers, using the DNA strand as template.
    The cycle is repeated, with the newly synthesised double stranded DNA being heat-denatured and the enzymes extending the primers attached to the liberated single DNA strands.
    The chain reaction, once set up, results in the exponential amplification of the original DNA, where the number of cycles (n) determines how many copies of the DNA (2n) are produced.


    The applications of PCR are extensive, some are:

    Amplification of small amounts of DNA for further analysis by DNA fingerprinting.
    The analysis of ancient DNA from fossils.
    Mapping the human (and other species) genome.
    The isolation of a particular gene of interest from a tissue sample.
    Detection of microorganisms present in low numbers in soil, food or water.

      احب اضيف ان اغلب القسام بدات باستخدامه .....سواء كانت ميكروبيولوجي , سرولوجي , هيماتولوجي , و طبعا اكيد في الموليكيلر ....


      الانترنت غني بشرحه و لا يسعني هنا سوى وضع نبذه عنه من باب العلم بالشئ ليس الا ....


      اتمنى ان اكون قد افدتكم واي سؤال انا حاضره ...... و لو عندكم اي سؤال هذا منتدى وضعته شركه متخصصه في الاجابه على استفسار .....

          في فقرة لو غيرتيها كان افضل..(تكبير وتكثير..)

            اخي الفاضل the forget ....... لقد عنيت قاصده ان اضع (تكبير و تكثير ) ولا تنساني حين يقولها لكم الدكتور جمال السعيد بهذا اللفلظ الذي لن تنساه ابدا ............... اتمنى ان المعلومه وصلت ..


            و لو احببت ان اضع لها فلا رديف لها سوى كلمه Amplification .......
            اما المفردات التي حضرتك ذكرتها فهي تطلق على الحاله المرضيه للمريض للعضو المصاب ولا تستخدم في غير ذلك الموضع (خارج جسم الانسان) ...

            و دمتم .

                وبدأ يستخدم في كثير من الابات في اكتشاف الاورقانيزمز مبكرا

                        PCR

                        Assalamu alaikom,
                        Thank you very much for this nice demonstration for this technique...
                        and i wanna say that this technique is very easy even you are beginner....U just have to follow a specific protocol......and dont be afraid of technique because its widely used.....PCR reaction mixture takes 10 minutes to prepare...and just run in the machine...
                        Now there are alot of PCR types...and more uses for them....
                        Anyway..what i wanna say for the students...that each PCR kit....has a manual according to the company that produced this kit....so you have to follow different protocols when dealing with different kits...but its the same principle....

                        So dont be scared of such simple technique...
