    هلا والله اخوي

    بصراحه والي اتذكره ان نزرع الماء بنقطه منه بعد المكس الجيد
    ونعمل ستريك بطول البليت وبعدين بالعرض وبعدين بشكل مائل علشان يكون السترينق الى البليت كامل ...ونشوف بعدين الكولوني الي تطلع ونحسبه ...بس بصرحه ماتذكر كم يكون Significant ??

    ياشباب ويا صبايا الا لسه يدرس معلوماته وفيره ...ساعدونا الله يساعدكم

      اخي اهم الفحوصات الخاصه بلمياه اذا كانت عذبه:
      1- العدالكلي القياسي :T.P.C.بواسطه بيئه nutrent agar حيث تاخذ واحد مل الى طبق بتري ثم يصب عليه البيئه ويترك الى ان يتصلب افضل قيمه للعدهي100/1ml
      2-البحث عن البكتريا البرازيه coliform وهذا تزرع بطريتين اما بطريقه ه الصب او العد الاكثر احتمالا على بيئه الاولى ماكوني اجار والثانيه ماكوني بروث مع انبوبه درهام
      3- بكتريا البيسدوموناس ارجونوزنا على بيئه سيرا ميدىتعطي رائحه العسل
      واذا تريد المزيد تواصل معي

        نفس الطرريقه تعمل عندنا لكن على blood agar
        وبعدين نبدأ العد
        انا حابه اسأل عن طريقة تجميع عينات الماء سواء
        dialysis or enviromental

            اريد ان اعرف العد الاكثر احتمالا بطريقة الثلاث انابيب وخمس انابيب وسبع انابيب

              يتم البحث عن بكتيريا القولون ايضا بطريقة الاختبارات الثلاثة 1- الاختبار الاحتمالي باستخدام بيئة الماكونكي السائلة

              2-الاختبار التاكيدي باستخدام بيئة الماكونكي اجار

              3-الاختبار التكميلي باستخدام بيئة EMB

              ثم تجرى اختبارات اخرى لتمييز نوع بكبيريا القولون

                على ماأعتقد ان العدد بكتريا الE.coli عن 10 بكتريا لكل 100 مل ماء.

                  السلام عليكم
                  الموضوع ليس بالهين وليس بكلمتين ممكن نقوله الموضوع كبير وهو كالتالي فالماء يعتبر عنصر هام لنا كما نعلم ولذا لابد من الاهتمام به ومعرفة كيف نحفظه من وجود الكائنات العديده من بكتيريا وفيروسات وطفيليات وفطريات وغيرها فهناك العديد من الامراض التي تنتقل عبر المياه
                  Waterborne diseases are caused by pathogenic microorganisms which are directly transmitted when contaminated drinking water is consumed.
                  Waterborne disease can be caused by protozoa, viruses, bacteria, and intestinal parasites.
                  واليكم بعض الامثلة للامراض مثل
                  Protozoal infections: Amoebiasis (hand-to-mouth)
                  Parasitic infections: Schistosomiasis (immersion)
                  Bacterial infections: • Botulism .
                  • Campylobacteriosis.
                  • Cholera .
                  • Chronic granulomatous disease - caused by the Mycobacterium marinum infection and localized in
                  • Diarrheal disease due to E. coli.
                  • Dysentery - Shigella/Salmonella bacteria - gastro-intestinal food/water.
                  Viral infections: • Adenovirus infection .
                  • Astroviruses.
                  • Caliciviruses.

                  وقبل فحص المياه هناك امور لابد ان نعلمها وهي:
                  Water Samples for Bacteriological Analysis
                  Containers:
                  1- Glass bottles, wide mouth, ground glass stoppers, with external protection.
                  2- The capacity should be at least 200 ml.
                  3- They should be sterilized by autoclave or Hot Air Oven.
                  4- A sufficient amount of Sodium thiosulphate should be added to the containers for collection of chlorinated water, to prevent continuation of bactericidal action during transit, before sterilization.
                  5- Plastic bottles are used for shipment of samples.
                  Collection procedure:-
                  General precautions in collection of samples:-
                  1- The samples should be representative to the water being examined.
                  A- Sea water samples
                  a) Samples should be taken from the most crowded area with swimmers.
                  b) Samples should be taken from a depth of 3 feet (chest level).
                  c) Samples should be taken from different sites and at different times.
                  d) The sample bottle is plunged downward below the surface with the mouth against the wave movement.
                  B- River water samples:-
                  a) Samples should be taken to a depth about 1 feet (30 cm) below the surface or to half the depth if it is less than 30 cm.
                  b) Samples should be taken from different sites across the river.
                  C- Swimming pool water samples:-
                  a) Samples should be taken from the 4 corners of the swimming pool at a suitable depth.
                  b) Also a surface sample is collected, where an oily layer from bather’s skin accumulates.
                  D- Tap water samples:-
                  a) Any attachment to the tap should be removed (rubber tube).
                  b) The nozzle of the tap and surrounding surface should be cleaned with a piece of cloth, sterilized with a Bunsen flame for one minute.
                  c) The tap is turned on, and the water is let to run for 2-3 minutes to clean any stagnant particles in the pipes. Then collect water sample.
                  E- Bottled water:-
                  The bottled water should be collected sealed and with a sheet containing the following information: Brand name, bottle volume, production date, validity period, the condition upon purchasing (refrigerated or not) and the place of purchase.
                  Prior to bacteriological examination, each sealed bottle is vigorously shaken 25 times by inverting upside down. Then it is opened and one quarter of its content is discarded, recapped and shaked again as before to ensure homogenous distribution of the organisms in the water sample.
                  F- Wells water samples:-
                  a) Wells with hand pumps. Operate the hand pump for about 5 minutes ( to waste stagnant surface water), then collect the sample.
                  b) Wells with mechanical pumps. The water is collected from a tap on the discharge.
                  c) Wells with no pumping machinery. The sample should be collected directly from the well by a bottle fitted with a weight or by a special sampler
                  2- The amount of water should be sufficient, should not be less than 100 ml. in search for pathogens, the sample volume may be 500 ml or more.
                  3- The bottle should be kept unopened till the amount of filling.
                  4- The bottle is held at the base in one hand, while the other hand is used to remove the stopper and cover together. While filling the bottle, the stopper and cover should kept in the hand. After collection the stopper and cover should be replaced immediately.
                  5- The bottle should not be filled completely; an ample air space must be left.
                  6- When several samples are collected from the same source on the same occasion, the sample for bacteriological examination should be taken first.
                  7- The sample should be clearly labeled and accompanied with a form ( card) containing detailed information e.g. date of collection, sampling site, sampling point, if chlorinated or not, preserved or not, any C/0 about odor and taste.
                  8- The sample should be sent to the laboratory as soon as possible, if delay is expected, the bottles should be kept in an ice –box.
                  9- In the laboratory, the samples are either processed immediately or kept in refrigerator for not more than tow hours.
                  Sample mailing
                  1- During mailing, efforts should be done to never send the sample in holidays.
                  2- Postal regulations should be considered as:
                  a) The kind of information.
                  b) The mail box. It is a single container with sufficient internal packing.
                  c) Plastic bottles are used.
