CROSSMATCH PROCEDURE-GEL METHOD
The crossmatch compatibility test is used to detect the presence of blood group antibodies in an intended
recipient’s serum/plasma directed toward antigens present on donor red blood cells.
PROCEDURE
1- Label a test tube for each donor to be tested
2- Dispense 0.5mL of Diluent 2 into the labeled tubes.
3- add 10uL of the donor packed red blood cells from the segment to the tube of Diluent 2 and Mix gently.
4- Label the gel Card with the appropriate identification and test information
5- add 50uL of each donor unit 0.8% red blood cell suspension to the correct microtube. Do not touch pipette to gel card.
6- add 25uL of patient serum or plasma to the correct microtubes.
7- Incubate at 37° +/- 2° C for 15 minutes
8- Centrifuge the gel card for 10 minutes.
9- Read the front and the back of each microtube
