What is microerophilic bacteria
specifice types of bacteria require oxygen to surviving but requires enviroments containig lower levels of oxygen than are present in the atmosphere

- suspend the required amount of sample or its rins in a suitable amount of diluted buffer
- centrifuge at 650-800 rpm for 10 minutes
- collect the supernatant
- pass the supernatant through two 25 mm filter chamber
aded 2-4 drops of the filtrate are spread on chocolate agar and incubated under microaerophilic enviroment
b-)selective Broth Enrichment and plating
Doyle and Roman enrichment broth consist of Brucella broth with added antibiotic suplement is inoculated with 10 to 25 gram sample,and incubated with agitation under microaerophilic atmospher at 42C for 16-18 hours
the enrichment culture is plated on selective media, and the plates are incubated at the same condition as an enrichment broth with no agitation required
